Details
Ultrasolute™ Amphipol 18 from Cube Biotech is a highly purified copolymer of cyclooctyl acrylamide and acrylic acid, with a molecular weight (Mw) of ~7.300. After dissolving, the copolymer is in a concentration from 1.0 to 5.0%. Copolymers provide a hydrophobic surface facing the membrane phospholipids and a hydrophilic surface at the hydrophilic outside. This setup makes nanodiscs highly soluble in aqueous solutions and allows for the solubilization of membrane proteins in the absence of detergents. As a secondary possibility the products can be used to creat 100% artificial nanodiscs. Phospholipids such as dimyristoyl-glycero-phosphocholine (DMPC) or palmitoyl-oleoyl-phosphatidyl-choline (POPC) have been proven to work here in combination with sodium cholate. The complex of Ultrasolute™ Amphipol 18 and membrane protein can be used in many biophysical assays, such as SDS-PAGE, SEC, Western Blot, UV/Vis spectroscopy, and many chromatographic procedures. Ultrasolute™ Amphipol 18 is part of our Synthetic Nanodisc Screening Kit MAXI. Together with 16 other nanodisc polymers you can screen for the best one for your membrane protein.
